Argonne Cancer Research Hospital Argonne National Laboratory Los Alamos Scientific Laboratory Paci f i c Northwest Laboratory S1 oan-Ketteri n g Inst. for Cancer Research Univ. California, eerkeley Univ. California, Los Angeles Short Description: During the period 1950-1980 many tracer studies were conducted on human subjects f o r the ‘purposes of studying the physiology and metabol ism o f labeled substances. Usual l y only a few subjects are invol ved No late effects are expected and i n general, there has not Brief description of the studies are: (1) Progesterone-4 1abel ed with carbon-14 was used t o study progesterone metabolism i n two patients. who was 10 weeks .pregnant was given 28.3 uCi. ‘Tter because of severe sickle cell anemia was performed. Activity i n ;ernal and fetal tissues studied. Storage of the hormone i n the fat Doses were 5.27 and 37.81 uCi. A third patient A therapeutic abortion 6 days rtments were found. (1958.)

pestigate whether synthesis o f Bence-Jones protein is related to that of (3) Hetabolism o f glucose studied with C-14, by CO-2 in expired air samples. Urinary and ex pira tory CO-2 were measured. (1958.) (4)Sequential studies were made ofthe body potassium content of man i n various cardiac disease states, i n a1 do ster on i sm, and i n hypertension with suspected hyperaldosteronism. A special study was made o f the re1ation ship between p l asma potassium shifts and total - body content of potassium i n man in various disease states. They are also measuring the effect of magnesium deficiency on 1 eve1 sof sodi urn, pot as si urn, magnesi um, and calci um inpl asma, cerebrospinal fluid, brain and skeletal nuscle. (1968.) (5) Retention curve data with Ca-43, Sr-85, and radium were obtained. (1959.) (6) An zrm coy3tier was used t o measure activity in blood to determine the clearance of I - Rose Bengal i n the 1iver. The curve has two exponential (7, 8 and 15) (1965.) A study was conducted t o measure the long-term retention and localization of technetium i n humans following both oral and intravenous administration and to derive mathematical model s for excretion, which are very useful in determining the radiation dose t o humans from technetium isotopes in ' ' body. They are important from both medical and occupational standpoints. University of Washington Hospital in Sea ttl e. The subjects were hospital ized at the University of Washington Cl in ical Research Center for the first week. for their services. providing the equipment, making the whole body counting and excreta measurements and analyzing the data. AEC but not the University of Washington participation. (8) A study was conducted t o determine the uptake, retention, distribution and excretion of promethium i n humans following both oral and intravenous administration and t o study the effectiveness of DTPA i n removing promethium from the body. These concentrations were relevant to possible exposure o f 143PmC13 was administered to 14 volunteers by physicians of the Hanford Environmental Heal t h Foundation (HEHF). Who1 e body counting and excreta surements were made by PNL for one year following administration of the '"Pm. Six of the volunteers were a1 so injected with TA at various intervals before -and after the administration of the Pm. Both the HEHF and PNL contributions to the study were funded by AEC. :'Tc and 96Tc were administered to8subjects by physicians at the Whole body counting and excreta measurements were made for The University of Washington obtained and reimbursed the subjects The participation by PNL personnel was limited to The PNL participation was funded by

' ( 9 a ) To extend the knowledge of tumor metabolism and t o define the l o c a l i - zation and improve the diagnosis of cancer i n animal models and man, studies have been i n progress over the seven year period (1977-1984). The specific aims are as follows: (1) To synthesize from different radiolabeled precursors a large variety of: (a) tj-13 and C - 1 1 labeled amino acids using enzymatic procedures. ( c ) l-(C-ll)-2-deoxy-D-glucose. (2) To empl oy the synthesized compounds f o r quantitative assessment of regional perfusion, transport, and metabol ism t o improve treatment of cancer, cancer diagnosis and the monitoring of treatment (9b) To investigate the a1 te ration in metabol i c patterns re1 ated t o cancer, methionine, technetium-99m and iodine-131 were used. These were used i n various compounds for the study of their metabolism i n bone, parathyroid tissue,1i ver, 1ung, pancreas and brain. Research was directed towards basic metabol i c patterns and their a1 te ration by mal ignant processes and by therapy, and also towards the development of useful diagnostic tests for early malig- (10) Various hematological studies included: (1959.) a) i n 35 patients iron kinetics with Fe-59 and cell volume with P-32 b) survival of red cell s, white cells and platelets with P-32 c ) intermediary metabolism of glucose and uric acid with C-14 glucose and C-14 uric acid. d ) heart function with 1-131 albumin. (11) The metabolic turnover of zinc-65 was studied, using a total body (12) a ) 1-131 Hip puran was compared with other renogram agents and found t o have the h i gher renal excretion efficiency .b ) 1-131 cholegrafin was used i n 35 patients to measure plasma volume as a substitute for human serum albumin. c) 1-131 Rose Bengal was used t o study hepatic blood flow compared w i t h d) Sr-85 was used as a tracer for calcium metabolism. Sr-90 and more available than Ca-47. (1959.) This is safer than (13) A radioisotope technique for measuring regional blood flow was utilized t o measure the effect of ionizing r.adiation on the bowel i n experimental studied i n women receiving radiation therapy for carcinoma of the cervix. They were studied before radiation, immediately a f t e r radiation and 6-12 weeks after radiation when healing had occurred. diseases of the col on such as ulcer at i ve col it i s and divert i cul it is were investigated to find out i f there were detectable disturbances i n regional In humans the e f f e c t of ionizing radiation on the colon was In addition, other

To evaluate the mechani sms control ring pulmonary circul ation, studies w r e carried out in normal and pathologic subjects, i n basal conditions and under various stimul ations. T h e data confirmed the re1 ationships previously found between pulmonary blood vol me, stroke vol ume and total blood vol ume and provided new material f o r an integrated evaluation o f mechanisms controlling the pulmonary circulation. (1960-1965.)
